mCerulean EVs are bioengineered EVs, loaded with mCerulean cyan fluorescent protein in their lumen using Ciloa’s pilot peptide technology, which does not overexpress surface proteins.
mCerulean EVs are produced in human embryonic kidney (HEK) 293T stable cell line and purified according to in-EV purification process, combining Tangential Flow Filtration and chromatography. Pure mCerulean EVs are subjected to standardised quality controls showing the total protein content, the number of particles, the EV size distribution, the presence of internal EV markers and the absence of cell protein contaminants. CD81 surface marker presence is revealed by a direct ELISA on pure mCerulean EVs. Cellular expression of mCerulean, its sorting within EVs and fluorescence measurement are given.
o Purity: in-EV standardised purification process combining Tangential Flow Filtration (TFF) and chromatography allows to obtain highly purified EVs.
o Quality: mCerulean EVs have been characterised using in-EV standardised quality controls.
o Storage: mCerulean EVs are in a simple buffer and are stable at +4°C for several months.

o Many applications: mCerulean EVs can be used as an internal reference for data normalisation, EV instrument calibration or as a positive control for EV quality control in cell fluorescence imaging.
