Nluc EVs, the new stable luminescent generation of EVs

Nluc EVs are produced in human embryonic kidney (HEK) 293T stable cell line and purified according to in-EV purification process, combining Tangential Flow Filtration and chromatography. Pure Nluc EVs are subjected to standardised quality controls showing the total protein content, the number of particles, the EV size distribution, the presence of internal EV markers and the absence of cell protein contaminants. A direct ELISA on pure Nluc EVs reveals the CD81 surface marker. Cellular expression of Nluc, its sorting within EVs, and luminescent measurement are given. EV luminal localisation of Nluc is demonstrated.

Strengths of Nluc EVs

o   Purity: in-EV standardised purification process combining Tangential Flow Filtration (TFF) and chromatography allows to obtain highly purified EVs.

o   Quality: Nluc EVs have been characterised using in-EV standardised quality controls.

o   Storage: Nluc EVs are in a simple buffer and are stable at +4°C for several months.

o   Performance: Nluc EVs are stably-luminescent EVs.

o   Many applications: Nluc EVs can be used as an internal standard to assess the effectiveness of EV isolation methods, to evaluate EV purification performance, in cell capture assays or in vivo for biodistribution studies.
