Quantitative analysis of EV recovery from conditioned cell culture media using EV-Down CM kit

in-EV is delighted to share the results of its collaborative study with BMG LABTECH, highlighting i) the interest of Nluc EVs as tracers for EV isolation and ii) the performance of EV-Down CM kit to isolate EVs from conditioned culture media.

Methodology

  • Use of bioengineered EVs, loaded with the NanoLuc® luciferase, as luminescent tracers
  • Luminescence quantification performed using the CLARIOstar® Plus high sensitivity multimode plate reader
  • Comparison of EV recovery after 2 h or overnight incubation
  • EV isolation performed on several culture media

Results

  • Efficient recovery of EVs using EV-Down CM kit
  • Stable luminescent signal in accordance with the expected concentrations
  • High reproducibility

       > EV-Down CM kit is a simple, efficient and rapid solution (without ultracentrifugation) for researchers isolating EVs from conditioned culture media

       > Nluc EVs are powerful tracers for tracking EVs during isolation steps and to determine recovery rates
